ABSTRACT

Objective:To explore the relationship between endocannabinoid (eCB) and its metabolic enzymes and severity of autism spectrum disorder (ASD), and to provide a theoretical basis for the study of the etiology and pathogenesis of ASD.Methods:A case-control study was conducted to collect 58 ASD children who underwent rehabilitation training at the Children's Developmental Behavior Research Center of Harbin Medical University and the provincial autism rehabilitation facility from December 2017 to December 2018 as the ASD group.According to the principle of gender and age 1∶1 matching, 58 normal children were selected as control group in Heilongjiang Province.Liquid chromatography-tandem mass spectrometry (LC-MS/MS) and real-time quantitative PCR (RT-qPCR) were used to detect eCB of ASD group and control group, including anandamide (AEA), 2-arachidonoylglycerol (2-AG), palmitoylethanolamide (PEA), oleoylethanolamide (OEA) and its metabolic enzymes: n-acylphosphatidylethanolamine-specific phospholipase D (NAPE-PLD), fatty acid amide hydrolase (FAAH), monoacylglycerol lipase (MAGL) and diacylglycerol lipase (DAGL) mRNA expression levels.Pearson correlation was used to analyze the level of eCB and ASD children's severity.Results:The levels of AEA, OEA and PEA in ASD children ((10.10±2.6)nmol/L, (24.30±5.60)nmol/L, (15.92±2.28)nmol/L) were lower than those in the control group ((13.46±3.04)nmol/L, (27.85± 6.89)nmol/L, (17.87±2.67)nmol/L, t=-6.612, -3.99, -4.779, P<0.01). The expression levels of FAAH and DAGL mRNA in ASD children were significantly higher than those in the control group, and the difference was statistically significant( t=2.423, 3.840, P<0.05), while NAPE-PLD and MAGL mRNA levels were not significantly different between the two groups ( t=0.024, 0.885, P>0.05). The level of PEA in the ASD group was negatively correlated with the total score of the autism behavior checklist (ABC) ( r=-0.288, P<0.05). Conclusion:There may be metabolic abnormalities in eCB and its metabolic enzymes in ASD children, and the level of eCB is related with the severity of ASD.

ABSTRACT

@#Objective To observe the effects of different health education patterns on chronic nonspecific low back pain (CNLBP). Methods From September, 2016 to April, 2017, 75 patients with CNLBP after rehabilitation were randomly divided into control group (n=45) and Back School group (n=30). The Back School group received group teaching including physiological and anatomy of lumbar spine, ergonomics, healthy posture and function exercise, once a week for four weeks. While the control group received the booklets including the same contents. They were self-assessed with Visual Analogue Scale (VAS), Oswestry Disability Index (ODI), and the 36-item Short Form (SF-36) before education, and one and three months of follow-up in clinics, or with call back services and Wechat. The recur-rence frequency was recorded. Results The score of VAS increased in both groups in the follow-up, but increased less in the Back School group (Z>2.645, P<0.01). The recurrence frequency was less in the Back School group (Z=-2.082, P<0.05), with more ODI score (Z=2.265, P<0.05) after three months of follow-up. The bodily pain score of SF-36 was more in the Back School group after one and three months of follow-up (t>2.273, P<0.05). Conclusion Health education with Back School may benefit to maintain the curative effects and function, reduce recur-rence, and improve the quality of life of CNLBP patients.

ABSTRACT

· AIM:To investigate the correlation between retinal thickness (CSRT) in the macular region and glycosylated hemoglobin (HbA1c) of patients with type 2 diabetes mellitus.· METHODS:Totally 39 cases of patients with diabetes (77 eyes) who screened from May 2016 to March 2017 were selected,and were divided into two groups according to the levels of HbA1c,which the 24 cases (47 eyes) in the low HbA1c group (HbA1c<8%) and 15 cases (30 eyes) in high HbA1c group (HbA1c≥8%).Other 22 cases of normal people (normal control group) and who for healthy physical examination were selected in the same period.Then,the correlation between HbA1c level and CSRT were analyzed by the Spearman correlation analysis.· RESULTS:In the high HbA1c group,HbA1c was (10.45±1.30)%,FBG was 10.67±1.64mmol/L and 2hPG was 15.98± 1.38mmol/L,which was higher than that in the low HbA1c group,and there was significant difference between the two groups (P< 0.05).The CSRT in the normal group was lower than the low HbA1c group and the high HbA1c group,and there was significant difference between the groups (P< 0.05).According to the analysis of the Spearman method,there was positive correlation between HbA1c and CSRT,macular volume,average macular thickness,FBG and those parameters,2hPG and them (P<0.01).· CONCLUSION:The HbA1c level is associated with retinal thickening in the macular region of patients with diabetes,which could predict the severity of diabetic retinopathy and provide important guidance for prevention and treatment.

ABSTRACT

Objective@#To observe the effects of swimming plus medication on the expressions of cytokines in rats with chronic abacterial prostatitis (CAP).@*METHODS@#Forty healthy adult male SD rats were randomly divided into five groups of equal number, normal control, CAP model control, medication, exercise therapy, and exercise + medication. The CAP model was made by Xiaozhiling injection, and at 7 days after modeling, the rats in the medication and exercise + medication groups were treated intragastrically with Qianlie Shutong Capsules (0.016 g/ml) at 20 ml per kg of the body weight qd, those in the exercise therapy and exercise + medication groups were made swim at a regular time once a day, 35 minutes on the first day and 5 minutes more on the second until 50 minutes once, for 4 successive weeks, and those in the normal control, model control and exercise therapy groups received normal saline only. After 14 and 28 days of treatment, all the rats were killed and their prostates harvested for observation of histopathological changes and determination of the expressions of TNF- α, IL-1β and IL-6 in the prostatic tissue homogenate by ELISA.@*RESULTS@#After 14 days of treatment, the expression levels of TNF-α, IL-1β and IL-6 were significantly elevated in the groups of CAP model control ([183.08±8.07] pg/ml, [57.55±3.53] pg/ml and [256.15±13.95] ng/L), medication ([118.49±8.06] pg/ml, [42.64±4.64 ] pg/ml and [200.74±9.33] ng/L), exercise therapy ([169.63±10.64] pg/ml, [50.45±5.71] pg/ml and [245.23±6.49] ng/L), and exercise + medication ([107.82±7.81] pg/ml, [40.35±6.93] pg/ml and [187.04±10.85] ng/L) as compared with those in the normal control ([20.36±1.82] pg/ml, [14.64±1.91] pg/ml and [70.58±2.09] ng/L) (P<0.05). At 28 days, the levels of TNF- α, IL-1β, IL-6 were remarkably lower in the exercise + medication group ([29.30±3.78] pg/ml, [16.91±1.24] pg/ml and [ 88.65±6.74] ng/L) than in the medication group ([39.67±3.19] pg/ml, [26.27±3.49] pg/ml and [110.26±6.33] ng/L) (P<0.05) and close to those of the normal control group ([19.34±1.76] pg/ml, [13.68±1.06] pg/ml and [71.34±2.50] ng/L). During the treatment, no obvious pathological changes were found in the prostate tissue of the normal control rats, while significant chronic prostatic inflammation was observed in the CAP models, and the inflammation was relieved in different degrees after intervention, most significantly in the exercise + medication group.@*CONCLUSIONS@#Swimming can relieve prostatic inflammation and swimming plus medication can effectively reduce the expressions of cytokines and alleviate histological damage in the prostatic tissue of CAP rats.

ABSTRACT

AIM:To investigate the effect of cepharanthine on the growth of human lung carcinoma A549 cells and the underlying mechanism. METHODS:After A549 cells were treated with cepharanthine,the growth inhibitory rate was detected by MTT assay. The cell morphological changes were observed under light microscope. The apoptosis of the A549 cells was analyzed by flow cytometry. The expression of microRNA (miR)-150, miR-182, p53 mRNA and FOXO1 mRNA were detected by real-time PCR. The downstream target genes were predicted by software,and the expression of p53 and FOXO1 was determined by Western blot. RESULTS:After cepharanthine treatment,the growth of A549 cells was in-hibited,the apoptosis rate was significantly increased,and the expression levels of miR-150 and miR-182 were significantly decreased. With cepharanthine treatment at 10 μmol/L,the expression levels of p53 and FOXO1 were elevated;however, with cepharanthine at 30 μmol/L,the expression levels of p53 and FOXO1 were decreased. After transfection with miR-150,the expression of p53 was significantly decreased, while the expression of FOXO1 was significantly decreased after transfection with miR-182. CONCLUSION:Cepharanthine inhibits the growth of A549 cells and promotes the apoptosis of A549 cells by inhibiting the expression of miR-150 and miR-182. miR-150 and miR-182 may down-regulate the expression of p53 and FOXO1,respectively.

ABSTRACT

Objective To analyze the status and trends of soil-transmitted nematode infections in Jiangxi Province from 1989 to 2014,so as to provide the evidence for generating the strategy of soil-transmitted nematode prevention and control. Methods The data of three epidemiological surveys on human parasitic diseases(in 1989,2002 and 2014)were classified and analyzed. The stool examination by Kato-Katz's thick smear method was adopted for the investigation of soil-transmitted nematode infec-tions. Results The total infection rate of soil-transmitted nematodes decreased by 91.89%from 77.67%in 1989 to 6.30%in 2014,in which the infection rate of Ascaris lumbricoides decreased by 98.78%from 71.11%to 0.87%,the infection rate of Trich-uris trichiura decreased by 96.80%from 29.67%to 0.95%,and the infection rate of hookworm declined by 73.57%from 17.63%to 4.66%. The infection rates of soil-transmitted nematodes in the female were higher than those in the male in three surveys. In different ecological districts,the infection rates of soil-transmitted nematodes in the female were also higher than those in the male,except in Zhe-Min Ecological District in 2002 and 2014. A declined trend of the infection was showed in all age-groups in the three surveys,but it slowed down by the growth of age,i.e.,the reduction rate was 97.03%in the age group of<10 years while 80.62%in the age group of>70 years. In 2014,the number of persons infected with soil-transmitted nematodes occupied 65.4%of the whole number of persons infected with intestinal parasites. Conclusions The mean infection rates of soil-transmit-ted nematodes decrease obviously in human population in different ecological districts,but the soil-transmitted nematodes are still the main species in intestinal parasite infections. The sequence of dominant species changes from A. lumbricoides,hook-worm and T. trichiura in 1989 to hookworm,T. trichiura and A. lumbricoides in 2014. The rural female and elder people are the key population,while hookworm is the key species for the prevention and control of soil-transmitted nematodes.

ABSTRACT

This article reported the clinical features of one child with infantile hypophosphatasia (HPP) and his pedigree information. The proband was a 5-month-old boy with multiple skeletal dysplasia (koilosternia, bending deformity of both radii, and knock-knee deformity of both knees), feeding difficulty, reduction in body weight, developmental delay, recurrent pneumonia and respiratory failure, and a significant reduction in blood alkaline phosphatase. Among his parents, sister, uncle, and aunt (other family members did not cooperate with us in the examination), his parents and aunt had a slight reduction in alkaline phosphatase and his aunt had scoliosis; there were no other clinical phenotypes or abnormal laboratory testing results. His ALPL gene mutation came from c.228delG mutation in his mother and c.407G>A compound heterozygous mutation in his father. His aunt carried c.228delG mutation. The c.407G>A mutation had been reported as the pathogenic mutation of HPP, and c.228delG mutation was a novel pathogenic mutation. Hypophosphatasia is caused by ALPL gene mutation, and ALPL gene detection is an effective diagnostic method. This study expands the mutation spectrum of ALPL gene and provides a theoretical basis for genetic diagnosis of this disease.

ABSTRACT

<p><b>OBJECTIVE</b>To study polymorphisms of catechol-O-methyltransferase (COMT) and monoamine oxidase B (MAO-B) genes among Chinese patients with Parkinson's disease.</p><p><b>METHODS</b>Genotypes of the COMT and MAO-B genes of 1408 patients with Parkinson's disease was sequenced using Sanger method. And these patients were recruited by Chinese Parkinson Study Group from 29 research centers throughout the country.</p><p><b>RESULTS</b>The genotypic frequencies of COMT rs4680 AA, AG, GG were 8.9%, 42.0% and 49.1%. Those of rs4818 CC, CG, GG were 42.5%, 45.6% and 11.9%, respectively. The genotype frequencies of MAO-B rs1799836 A/AA, AG, G/GG were 74.4%, 14.1% and 11.5%, respectively. The haplotype formed by COMT rs4680 (GG) and MAO-B rs1799836 (A/AA) genotype has a frequency of 36.86%.</p><p><b>CONCLUSION</b>Polymorphisms of COMT and MAO-B genes has a unique characteristics among Chinese patients with Parkinson's disease. They may be related with differences in drug response in such patients.</p>

ABSTRACT

Objective To understand the status of mental health among left-behind children in rural area and to explore its relationship with social capital.Methods A sample of 1 031 leftbehind children,aged 10-18 years,was randomly selected from six schools of two townships in Macheng city of Hubei province where large number of rural parents were working in the urban area.Data from a cross-sectional survey was collected among the sampled students from primary,middle and high schools,using self-designed questionnaires which targeting the ‘ left-behind’ condition and social capital Mental Health Inventory of Middle-School Students (MMHI-60).Chi-square test was used to compare the differences in mental health among different groups.Spearman correlation and logistical regression analysis were applied to assess the relationship between social capital and mental health.Results The prevalence of mental problems was 54.12%.Results from linear x2 test indicated that the detection rates of mental problems were parallel with the monthly pocket money and the reduction in the frequency of contacting migrant father/mother (P<0.01).Results from the multiple logistic regression analysis indicated that students who were senior (OR=2.410-2.682,P<0.001),and who reported not caring about their parents (OR =2.048-2.420,P< 0.001) were at increased risks for mental health problems.After controlling the demographic characteristics and conditions of staying relevant factors,family social capital (OR =0.845,95% CI:0.801-0.891),neighborhood social capital (OR=0.867,95%CI:0.826-0.910) and school social capital (OR=0.893,95%CI:0.863-0.923) were all associated with declined risks of mental health problems,in which the family social capital was the most important one.Conclusion The prevalence of mental health problems was high among the left-behind children in rural area.Much attention should be paid to assist these children in improving their mental health status.One promising approach appeared to be improving social capitals which would include family social capital,neighborhood social capital,and school social capital to assist these children from avoiding mental health problems.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the 4-hydroxynonenal (4-HNE) expression changes and the impact of ulinastatin (UTI) METHODS: Seventy-two healthy Sprague-Dawley rats were randomly divided into three groups: the control group, poisoning group and treatment group, with 24 rats in each group. The model of lung injury was established by intragastric PQ (80 mg/kg) administration in poisoning group and treatment group, and 1 mL saline was administered intragastrically in the control group. The rats in treatment group were injected intraperitoneally with UTI (100 000 U/kg) 30 minutes after PQ administration, and the rats in the control group and poisoning group were intraperitoneally injected with the same volume of saline. After different treatments, the pathological changes and the expression of 4-HNE in lung tissue was detected in 12, 24, and 72 h in three groups.</p><p><b>RESULTS</b>In the poisoning group and treatment group, the expression of 4-HNE in lung tissue of rats were increased in 12 h after poisoning and reached the peak in 48 h; in 72 h after poisoning, the expression of 4-HNE in lung tissue were decreased, but they were still high. Compared with the control group, the expression of 4-HNE in lung tissue of rats were significantly increased in the poisoning group and treatment group (P < 0.05). And compared with the poisoning group, the expression of 4-HNE in lung tissue of rats were significantly decreased in the treatment group (P < 0.01). The pathological changes were observed, including alveolar capillary expansion, diffuse alveolar hemorrhage and alveolar inflammation cell infiltration, were found in lungs of rats in poisoning group and treatment group. There is no significant change in the control group. Compared with the control group, the expression of 4-HNE in lung tissue significantly increased in poisoning group and treatment group (P < 0.01), but the expression in treatment group was lower than in poisoning group (P < 0.01).</p><p><b>CONCLUSION</b>The expression of 4-HNE increased in PQ intoxicated rats. UTI may reduce the expression of 4-HNE and reduce lung injury in PQ intoxicated rats.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To trace the embryonic stem (ES) cells transplanted into rat brain by labeling the cells with green fluorescent protein (GFP) and by mouse neuronal specific antibody Thy-1 and compare their features.</p><p><b>METHODS</b>For GFP labeling,transfect pEGFP-N1 plasmid containing GFP and anti-neomycin sequences into embryonic stem cell and add neomycin for more than 10 passages. To test the GFP expression in vivo, the GFP-ES was transplanted into healthy rat brain, and the frozen sectioned slides were observed under fluorescence microscope and laser con-focal microscope 21 days later. For the antibody labeling,embryonic stem cells were directly transplanted into the rat brain. The specific mouse thy-1 antibody was used in immunostaining of transplanted cells. For both of the two labeling method, the slides were also examined by double labeling with the antibodies,neuronal nuclei (NeuN) or glial fibrillary acidic protein (GFAP) to identify the differentiation of transplanted cells.</p><p><b>RESULTS</b>Both single ES cell and cell pellets expressed bright green fluorescence the day after plasmid transfection, and more than 30% ES cells were labeled. The GFP-labeled cells could still be found gathered around the infusion channel at least 21 days later, but the GFP fluorescent could not be overlapped with NeuN or GFAP staining. On the contrary, Thy-1 antibody overlapped well with NeuN or GFAP staining.</p><p><b>CONCLUSIONS</b>Liposome-helped plasmid GPF transfection is effective in labeling mouse embryonic stem cell in vivo,but is not effective in showing the differentiated cells. On the contrary, Thy-1 antibody can not only show the transplanted cells, but also trace the transplanted cells after their differentiation.</p>

ABSTRACT

We transfected human neural stem cells using lentiviral vectors encoding glial cell line derived neurotrophic factor (GDNF) to study its expression level in vitro and to get a stable cell line expressing GDNF. First, GDNF gene was sub-cloned into the lentiviral transfer vectors. Then, the recombinant lentiviral supernatants were packaged by 293T cells through three plasmids transient co-transfection method using standard lipofectamine reagent. The viral titers were tested by the transfection efficiency of 293T cells. At the same time, human neural stem cells (hNSC) were transfected under different multiplicity of infection. GDNF gene expression level and protein secretion level of hNSC were tested by real-time PCR and ELISA methods after transfection. Lentiviral vectors encoding GDNF were constructed. Using lentiviral vectors encoding GDNF we successfully transfected human neural stem cells, and got a stable neural stem cell lines over-expressing GDNF. Furthermore, the results indicated that GDNF expression was influenced by the multiplicity of infection. Human neural stem cells could over-express GDNF through lentivial vectors tranfection. Its gene expression level and protein expression level correlate with the multiplicity of infection.
